"The exact scale of the show has not yet fully been agreed but it will be somewhere in the region of 30-35 acres," said a representative.
"We're working closely with the team at Chatsworth to ensure that the grounds are not adversely impacted upon. We anticipate some excavation activity and are working with experts and the Chatsworth team to agree the details and scope of this."
The RHS pulled out of BBC Gardeners' World Live (GWL) after the June 2015 show, but said it remains committed to gardening in the Midlands.
"We are no longer providing horticultural content for GWL, although for the foreseeable future RHS members will have the same discounted tickets to the show.
"As part of our £100m investment plans and commitment to increase our presence across the Midlands and north of England, the RHS's key focus is to create new RHS gardens and horticultural highlights in these parts of the country.
"On top of this, more than 3,000 schools in the Midlands are part of our Campaign for School Gardening and more than 500 gardening groups take part in RHS community campaigns including It's Your Neighbourhood as well as Britain in Bloom."
